What you're describing is using a chemical irritant, toothpaste often contains compounds that create a burning sensation in sensitive areas. This effect will vary greatly from person to person and also depending on the type of toothpaste you use.
Nipples are usually safe to experiment on with toothpaste, it will not harm you but be aware once the burning sensation starts there is nothing that will stop it instantly. This is common with all kinds of chemical type play, there is no "off switch" as the effect is due to the chemical being absorbed into your skin.
As mentioned by @subbysecret, things like Icy Hot / Deep Heat etc are a much harsher version of this and should only be attempted once you gain some understanding of how these things work on you.

Its much easier to do it with icy-hot or a similar product
you can also try using hot sauce
you can make your nipples tender by ripping nipple clamps off them then applying hot sauce or lemon juice
if you make a few small wounds on your nipples it will burn a lot more

A little extra tip the skin on the underside of the arm is relative sensitive while not that bad should there be a allergic reactions for example.
So I find it a good idea to test a small amount there first and see how you react to it before going to nipples or even more sensitive parts
